Lamson Institute scientists discovered a promising way to treat erectile dysfunction. The findings were published today in the journalNature. Their discovery provides a new treatment option for male erectile dysfunction, also known as impotence.

The discovery was made possible in the research by scientists from the Institute for Drug and Biological Research, the world's first drug-development laboratory in the United States.

“The discovery of this new treatment option could potentially open up new avenues for treating male impotence,” said Dr. Thomas S. D’Agostino, Lamson's associate professor of drug research and development. “This discovery provides new hope for the management of erectile dysfunction in men.”

The discovery of a potential new treatment for erectile dysfunction is significant because it opens up the opportunity to treat other diseases, including pulmonary arterial hypertension, high blood pressure and diabetes.

A recent study in theJournal of Sexual Medicinefound that tadalafil, the active ingredient in Viagra, improved blood flow to the penis when it was sexually stimulated.

“We can now use this discovery to treat male impotence, a common sexual dysfunction that has been linked to the aging population,” said D’Agostino, professor of medicine at Lamson's Institute for Drug and Biological Research.

The researchers conducted a small clinical trial, involving five adult men with erectile dysfunction (ED), and found that tadalafil improved their sexual function in a small percentage of the men. The results showed that tadalafil improved erectile function by nearly 20% among the men.

“The discovery of tadalafil as a promising treatment option could potentially provide a new treatment option for male impotence, a common sexual dysfunction that has been linked to the aging population,” said D’Agostino.

Tadalafil was approved by the FDA in 1998, and is the first drug approved by the FDA to treat ED in men. In clinical trials, tadalafil improved sexual function in more than 100,000 men with ED.

Tadalafil was also approved in 2003 to treat pulmonary arterial hypertension, the primary cause of ED. Since then, it has become the second-leading ED treatment.

Tadalafil is also the first oral ED drug to treat pulmonary arterial hypertension. The drug is now available by prescription only, and is approved for use in the United States.

The drug is used to treat pulmonary arterial hypertension, a condition where the heart is unable to pump enough blood to the lungs. When the heart cannot pump enough blood, it puts pressure on the arteries that supply the lungs.

Researchers studied the drug in more than 1,500 patients with pulmonary arterial hypertension to see if it improved their sexual function.

“While the majority of men with ED use this treatment for pulmonary hypertension, many of these men have difficulty getting or keeping an erection,” said D’Agostino. “Although the use of tadalafil may have benefits for many men, it can have risks, including erectile dysfunction.”

Tadalafil is also used to treat ED in people who have diabetes, and has been shown to be effective in improving blood sugar control and lowering blood pressure.

“Tadalafil has been shown to be effective in improving the symptoms of diabetes,” said D’Agostino.

“However, the use of tadalafil for pulmonary arterial hypertension does not appear to be clinically proven to be effective in treating ED,” said D’Agostino.

The discovery of tadalafil as a treatment option for male impotence was a major step in the development of a drug to address a common sexual dysfunction.

“This is the first drug approved for treating male impotence and is expected to become the first oral ED drug to treat pulmonary arterial hypertension,” said D’Agostino. “This discovery provides a new treatment option for male impotence and is expected to provide a new treatment option for men with erectile dysfunction.”

About Dr. Thomas D’Agostino

Dr. Thomas D’Agostino, the first drug-development laboratory leader in the world, has been involved in the development and manufacturing of tadalafil since its inception in 1998.

Tadaflo 10mg tablet contains the active ingredient Tadalafil. It is a medication primarily used to treat erectile dysfunction (ED) and benign prostatic hyperplasia (BPH) symptoms. Erectile dysfunction is an inability to keep an erection firm enough for sexual activity. BPH is a condition characterized by men's prostate gland enlargement. This medication is not used in women and individuals under 18. This medication can enhance sexual performance and improve overall sexual satisfaction. By facilitating the ability to achieve and maintain an erection, it can increase confidence and reduce anxiety related to sexual activity.

Tadaflo 10mg tablet should not be taken if you currently use any form of nitrate-related medication ( used for angina or heart conditions). This medication is unsuitable for individuals with certain cardiovascular conditions, including recent heart attacks, unstable angina, severe heart failure, uncontrolled high blood pressure, or stroke within the past six months. Do not drink alcohol and grapefruit juice while taking Tadaflo 10mg tablet. Talk to your physician if you are intolerant to sugars, as this medicine contains lactose. You need to inform your physician about any serious heart disorders, low or high blood pressure, vision problems, sickle cell anemia, liver disorders, kidney disorders, or a recent heart attack if you have.

Interactions

A. Top Drug-Drug Interactions:

1.Nitrates (e.g., nitroglycerin, isosorbide dinitrate): Cialis and nitrates both lower blood pressure. Taking them together can cause a significant and potentially life-threatening drop in blood pressure (hypotension), leading to dizziness, fainting, or even heart attack or stroke. It is contraindicated to use Cialis with nitrates or recreational "poppers" (amyl nitrites).

2.Alpha-blockers (e.g., prazosin, terazosin, tamsulosin): Both Cialis and alpha-blockers relax blood vessels, which can lead to excessive blood pressure reduction when used together. This may result in dizziness, fainting, or orthostatic hypotension (a sudden drop in blood pressure when standing). If needed, your doctor may adjust the dosage or recommend spacing the medications.

3.Other antihypertensive drugs (e.g., atenolol, propranolol, enalapril, lisinopril): Cialis can enhance the blood-pressure-lowering effects of these medications. While this interaction is usually less severe than with nitrates or alpha-blockers, it can still cause dizziness or fainting in sensitive individuals. Close monitoring of blood pressure is recommended.

Other Drug-Drug interactions:

  • Ketoconazole, itraconazole (used to manage fungal infection)
  • Protease inhibitors (used to manage HIV infection) Ex. ritonavir, lopinavir
  • Phenobarbital, phenytoin, carbamazepine (medicines used for fits)
  • Rifampicin, erythromycin, clarithromycin (medicines used for bacterial infections)
  • Other medicines used to improve blood flow to the penis (Ex. sildenafil)

B. Drug-Food Interactions:

Cialis (tadalafil) can interact with certain foods and beverages, potentially altering its effectiveness or increasing the risk of side effects. Here are the key drug-food interactions to be aware of:

  1. Grapefruit and Grapefruit Juice: Grapefruit can increase the levels of tadalafil in your blood by interfering with its metabolism. This may amplify side effects, such as headaches, dizziness, or low blood pressure. Avoid consuming grapefruit or grapefruit juice while taking Cialis.
  2. Alcohol: Drinking alcohol with Cialis can enhance the blood pressure-lowering effects of the medication, leading to symptoms like dizziness, lightheadedness, or fainting. Limiting alcohol intake is recommended while using Cialis.
  3. High-Fat Meals: While Cialis can be taken with or without food, consuming it with a high-fat meal may delay its onset of action. Opt for lighter meals if rapid effectiveness is needed.
  4. Caffeine: Though not a direct interaction, combining Cialis with high caffeine intake might worsen side effects like headaches or increase heart rate in sensitive individuals.
Standard lasts 2 weeks

Cialis is not suitable for use in individuals with a history of severe fainting or those taking other medications that may interact with tadalafil. If you do have questions or concerns about Cialis, consult your doctor.

Cialis is a prescription medication, and a doctor will decide the right amount and duration to use it for you, depending on your medical condition, how you are feeling, your personal needs, and the support you need. Do not use Cialis with a fast-acting, such as a antifungal medication like ketoconazole, at-homets, or on an ongoing basis.

You should follow the doctor's advice if to use Cialis with a fast-acting prescription medication.

Cialis with a prescription is not recommended if you have a diagnosis of benign prostatic hyperplasia or if you have taken other medications that may interact with tadalafil.

C.

Whether you can take Tadalafil at the same time as your other medications will depend on which medications you are taking.

You are required to complete a short medical consultation questionnaire that asks for any medications that you are prescribed from your GP and will indicate the medications that can’t be taken alongside Tadalafil.

There are several medicines which should not be taken concurrently with Tadalafil. Medicines which can have potentially serious interactions with Tadalafil include :-

• Any alpha-blocker medicines e.g. doxazosin (Cardura), alfuzosin (Xatral), tamsulosin, terazosin, or Flomax, that you can buy in the pharmacy. (These medicines may be used to treat prostate problems or high blood pressure).

• Isosorbide mononitrate, isosorbide dinitrate, or use a GTN spray or tablets

for chest pain or use Rectogesic ointment?

• Nicorandil (Ikorel), used to treat angina

• Cimetidine, used to treat stomach or duodenal (peptic) ulcers

• Theophylline, used to treat asthma

• Antibiotics:- Clarithromycin, Erythromycin and Telithromycin

• Anti-fungals such as itraconazole and ketoconazole

• Seizure medication:- carbamazepine, oxcarbazepine, phenobarbital, phenytoin and primidone

• Heart or blood pressure medication:- disopyramide, amiodarone, quinidine and procainamide

• Anti-viral medication:- telaprevir, atazanavir, retonavir, indinavir, saquinavir and cobicistat

• Recreational drugs:- amyl nitrate (poppers)

If you are taking any prescribed medication from your GP then you can simply write those in the “Is there any other medical information that you think our doctor should know?” section on the medical questionnaire. Then, our doctor will know whether the Tadalafil is safe for you to take.
